Synthesis and Pharmacological Activities of Novel 3-(Isoxazol-3-yl)-quinazolin-4(3H)-one Derivatives

Several new 3-(isoxazol-3-yl)-quinazolin-4(3H)-one derivatives were synthesized and tested for their analgesic and antiinflammatory activities, as well as for their acute toxicity and ulcerogenic effect. A few compounds were as active as phenylbutazone in the writhing and acetic acid peritonitis tests. They had a very low ulcerogenic effect.

Conformationally restricted, orthogonally protected 2,4-diaminocarboxylates with a cyclopentane skeleton were efficiently synthesized from beta-lactam 6, the syntheses involving strategies of diastereoselective epoxidation of the beta-lactam and the corresponding monoprotected amino esters with opposite selectivities followed by regioselective opening of the oxirane ring with sodium azide. The enantiomers were also prepared. This new class of compounds can be regarded not only as conformationally constrained beta,gamma-diamino acid derivatives but also as potential functionalized carbocyclic nucleoside precursors.

Electrochemical and spectroscopic studies of poly(diethoxyphosphoryl)porphyrins

Abstract The synthesis and electrochemical characterization of two related series of porphyrins bearing diethoxyphosphoryl groups are reported. One group of compounds is represented as (T( p -R)PP)M where R = phos = P(O)(OEt) 2 and M = Zn(II) or H 2 while the other is represented as (di( p -R)Pdi(phos)P)M where R = P(O)(OEt) 2 , H or CH 3 and M = Zn(II) or H 2 . Each porphyrin was investigated by electrochemistry and thin-layer spectroelectrochemistry in CH 2 Cl 2 , CDCl 3 , CHCl 3 or PhCN containing tetra- n -butylammonium perchlorate (TBAP) as supporting electrolyte. Protection of all functional groups of the carbohydrate portion of the chemoenzymatically synthesized sialyl T threonine ester 1 (R=R1 =H, R2 =tBu, Fmoc=9-fluorenylmethoxycarbonyl) and subsequent acidolysis of the tert-butyl ester afforded the building block 2 (R=Ac, R1 =Me, R2 =H). The latter is a useful tool in the solid-phase synthesis of the N-terminal sequence 3 of the leukemia-associated leukosialin.

The role of Ag+ ions in the ultraviolet-driven photochemical synthesis of Au nanorods (NRs) in aqueous surfactant mixtures has been investigated in order to elucidate the mechanism that drives anisotropic nanoparticle growth. The α-alkylation of deprotonated N-aryl-α-aminonitriles with α-bromoesters furnishes intermediates that can be cyclized to 4-quinolones upon microwave irradiation. Alternatively, base-induced dehydrocyanation of the alkylation products furnishes enaminoesters, which can, for example, be converted into quinoline-3-carboxylates.

Starting from 2 and 3, obtained from santonin (1), we have synthesized natural guaianolides 4-7. Chemoselective epoxidation of 2 gave (+)-11betaH,13-dihydroestafiatin (4), and epoxidation of 3 followed by regioselective elimination of the hydroxyl group afforded (+)-11betaH,13-dihydroludartin (5). Sharpless' mild regioselective ring-opening of 4 and 5 followed by hydrogenolysis yielded (-)-compressanolide (6) and (-)-11betaH,13-dihydromicheliolide (7), respectively.

Synthesis and biological activity of new anti-inflammatory compounds containing the 1,4-benzodioxine and/or pyrrole system

A series of substituted derivatives containing the 1,4-benzodioxine or pyrrole nucleus are described. All the newly synthesized compounds were examined for their in vitro and in vivo anti-inflammatory activity. Several derivatives, including (S)-2, 14 and 17, showed more anti-inflammatory activity in vivo in these assays (rat paw oedema induced by carrageenan) than the known classical anti-inflammatory agent ibuprofen, whereas other compounds like 1 were equipotent to ibuprofen. The synthesis of metallocene-labelled drugs for biological assays

Several drugs (amphetamine, desipramine, nortriptyline, phenobarbital) have been labelled with metallocenic fragments in order to develop a new immunoassay method. The metallocenic fragments are cymantrenic or benchrotrenic derivatives: the linkage between the organic and organometallic moieties has been achieved by reactions between amino and acidic functional groups. All the products (metallohaptens), purified by different chromatography techniques, have been fully characterized by IR and 1H NMR spectroscopy and their mass spectra.
